Complement Activation

چکیده
The complement system is the most important biochemical cascade in the blood for the recognition, opsonization, and elimination of foreign materials. To date, the leading causes of death in the United States include cancer, cardiovascular and neurodegenerative diseases, and diabetes. New treatments are urgently needed to treat these devastating diseases and nanotechnology potentially provides new avenues to fi ght such illnesses. These avenues include the development of novel nanocarriers that deliver drugs in a specifi c and controlled manner, while minimizing secondary effects. The success of bioengineering effective nanocarriers for drug delivery purposes requires a deep understanding of the interaction between the complement system and the nanocarriers. This review focuses on reporting the current state of complement activation by different nanomaterials. Here, we assess various important parameters that infl uence the activation of the complement system, which include the physicochemical characteristics of both nanocarriers and complement proteins. We next evaluate the most recent engineering approaches to prevent or reduce complement activation. Finally, we discuss different in vitro and in vivo procedures to assess complement activation.

, Shaheed Beheshti University of Medical Sciences Abstract: Background: The main surgical side effects and complications in many cases are due to the long duration of the anesthesia. Obviously these phenomena are related to the use of anesthetic drugs which have effects on liver blood flow, immune system and homeostasis. One important effect is activation of complement system. In this stud...

Background: Coronary artery disease (CAD) is one of the most important and lethal diseases in the world. CAD represents a board spectrum of disease from silent ischemia at one end to sudden cardiac death at the other end. The middle of this spectrum consists of acute myocardial infarction (AMI) and unstable angina pectoris (UA). Recent data show that the inflammatory process plays a major r...

Background: Inappropriate activation or blockage of the inhibition of complement system could cause tissue damages in autoimmune diseases particularly rheumatoid arthritis (RA). Defect in complement component regulation may cause damages to tissues, on the other hand, or the damaged tissue might affect the unnecessary activation of complement components. Objective: To investigate the expressi...

Abstract Background and Objectives One of the side effects of immunoglobulin administration is the activation of the complement system related to the high concentration of aggregates/large polymers in the production process. Measuring anticomplementary activity (ACA) is important in pharmaceutical quality control and is a criterion of spontaneous activation of complement system in immunoglobuli...

Abstract Introduction: Thalassemia is the most common hereditary anemia in Iran. The aim of this study was to evaluate the humoral immune system and assess the effect of splenectomy on the serum level of immunoglobulins IgG, IgM, and IgA and complement components in patients with major thalassemia. Materials and Methods: This interventional semi-experimental study (before-after) was performed...
متن کاملContinual Low-Level Activation of the Classical Complement Pathway
There is evidence that the classical complement pathway may be activated via a "C1-tickover" mechanism, analogous to the C3-tickover of the alternative pathway. We have quantitated and characterized this pathway of complement activation. Analysis of freshly collected mouse and human plasma revealed that spontaneous C3 activation rapidly occurred with the generation of C3 fragments in the plasma...
